United face Championship side, Derby County managed by Frank Lampard, at Old Trafford on Tuesday.
The game will go straight to penalties if the scores are level after 90 minutes and because of the new rule, Mourinho has revealed he is “not thinking too much” about rotating his squad.
“I’m just thinking about the matches and this one is a cup match and the rules change.
“In this moment, you don’t even have the possibility of, after a draw, playing 30 more minutes — you go straight to a penalty shootout which obviously is a 50-50 situation.
“So it is a match of 90 minutes which we have to try and win and the choice of the players is based on that. We have to try and win,” Mourinho said, according to ESPN.
Mourinho has already confirmed that 19-year-old full-back, Diogo Dalot will start while Ander Herrera, Ashley Young, Scott McTominay, Eric Bailly, Anthony Martial and Juan Mata are all in need of games.
Nemanja Matic is available again after serving a one-match suspension.
